the ultimate reliable yeast two-hybrid screening technology

ULTImate Y2H™ is an optimized version of the yeast two-hybrid (Y2H) screening technology that tackles most of the false positives and false negatives issues arising from the original technology developed by Stanley Fields in 1989.

114 positive clones are delivered on average per screen and the sticky proteins are flagged.

Indeed, unlike classical Y2H™ screening performed by sequential transformation of a bait and then the library, ULTImate Y2H™ is derived from a unique and patented cell-to-cell mating process that allows the testing of 97 million interactions at once on average.

This screening power allows us to screen highly complex libraries exhaustively:
       > our random-primed cDNA libraries contain 10 million
          independent and primary clones,
       > a complexity considered to achieve comprehensiveness
          of the mRNAs present in a eukaryotic cell
                  → rare mRNAs are included
       > to be exhaustive, a screening should statistically achieve
          5x coverage of the library complexity
                  → we achieve 10x on average, don’t miss interactions!

Analysis of all positive clones combined to exhaustive screening makes our ULTImate Y2H™ screens 90%+ reproducible. Reproducibility allows us to compute a confidence score for each interaction that identifies non-specific interactions and potential false positives.

Sticky proteins are therefore flagged and you can work with trust on the most reliable interactions identified. Do not start working on artefacts!
