an optimized FRET screening assay

HTRF® is one of our solutions to develop a protein interaction assay suitable for High Throughput Screening .  This technology developed by Cisbio international is based on time resolved fluorescent energy transfer. Direct or indirect labelling of the protein interaction partners allows a quantitative detection of your favorite interaction. This assay is homogeneous and uses a small quantity of proteins.
We have developed numerous protein interaction assays with HTRF: intracellular interactions, proteases , and cofactors, viral protein-host protein, transcription factor-coactivator and apoptosis-related interactions.
Various tags for labelling and purification and a large set of buffer conditions are used to optimize protein interaction signal detection. Contact us for more details.

HTRF interaction assay principle.Antibodies bearing fluorescence donor (Europium cryptate, EuK) or acceptor (XL665) recognize the tags on each protein. Interaction between Protein1 and Protein2 is detected by fluorescence transfer (excitation at 337 nm, emission at 665nm). Emission at 620 nm occurs regardless of the interaction, and allows to normalize the assay.

HTRF protease assay principle. Antibodies bearing fluorescence donor (Europium cryptate, EuK) or acceptor (XL665) recognize both tags on the protease substrate. Substrate cleavage by the protease is detected by loss of fluorescence transfer (excitation at 337 nm, emission at 665nm). Emission at 620 nm occurs regardless of the cleavage, and allows to normalize the assay.
